Indomethacin is a non-steroidal drug with anti-inflammatory ac- tivity (NSAID), widely studied in man and laboratory animals in the 1970s. It has been used extensively in human medicine for the treatment of rheumatic diseases. Its capacity to inhibit the synthe- sis ofprostaglandins is well known (Ferreua et al., 1971). Studies of the kinetics of indomethacin in humans have shown that the disposition of this drug can be described by biexponential kinetics. Its distribution in the body is widespread: the drug crosses the placental barrier (Parks et al., 1977) and is excreted in milk (Takyi, 1970). It has a volume of distribution (V,,,,) in the wide range 0.3 - 1.6 L/kg and a plasma clearance (CllBl) of 0.1 1-0.8 L/h kg (Duggan etal.. 1972; Alvan et al., 1975). It is metabolized by demethylation, N-deacylation and glucuronide conjugation (Friedman et al., 1991), undergoing enterohepatic cycling (Yesair et al., 1970).
